Preclinical humanized cGAS mouse model

Design of the hcGAS mouse
The humanized cGAS model was developed by Knockin at the mouse cGAS locus and expresses a full human cGAS.

Validation
Human cGAS expression in brain and lung of cGAS humanized mice
cGAS expression was assessed in brain and lung of 9-week-old WT and hcGAS mice by Western Blot.
Human cGAS inhibitor prevents BCV-induced cytokine release in hcGAS, but not in WT mice
hcGAS and WT mice were treated with cGAS small molecule inhibitor or vehicle and, after 24h, were injected with different dosage of BCV. 6h later, plasma was harvested from the mice and cytokine levels (IFN-α, IP-10, IFN-β) were evaluated.
